After you have connected the Psion via the USB Comms link to your PC, and started Psi2Win, click the Setup button.
Usually only one of the listed com ports is green, and if so, choose that. If none of them are green, then the system must have chosen to use Com port with a higher number. Just enter 5 in the text box, and click the Check button to see if that works. If not, try 6, 7, etc until you get the right port.
If that is taking too long, and you are on a windows machine, you can open the Device Manager of windows (open the start menu and just start typing "Device manager" to search for it), and expand the Ports(COM&LPT) entry to see which port is being used.
